Submit ReviewPuromycin dihydrochloride is a protein synthesis inhibitor; leads to the premature release of polypeptide chains as polypeptidyl purine derivatives. Analog of the 3' end of aminoacyl-tRNA. Aminonucleoside antibiotic. Inhibits translation in both in vitro and in vivo systems. Also inhibits the transport of proteins into the mitochondria in vitro.

Puromycin (5 mcg/ml) along with geneticin (250 mcg/ml) was used in DMEM containing 10% FBS, 4 mM glutamine and 10 mM HEPES to maintain the stable expression of hRyR2-IRES-GFP in HEK293 cell culture. The stable expression of hRyR2 in these cells was confirmed by cytosolic GFP fluorescence and 10 mM caffeine-evoked cytosolic Ca2+ peak, measured by Fura2 ratiometric dye, as shown in figure.
Puromycin should be stored in aliquots at -20 degree C.
