Review excerpt: Mitogen-activated protein kinase (MAPK) networks are critical for the transmission of extracellular signals into appropriate intracellular responses. Prototypical MAPK activation employs a three-kinase core module consisting of a MAPK kinase kinase (MAPKKK or MAP3K) that phosphorylates and activates a MAPK kinase (MAP2K, MEK, or MKK) that in turn phosphorylates and dramatically increases the activity of one or more MAPKs.
