生物活性 for JQ dS-4

JQ dS-4 is a Brahma-related gene 1 (BRG1)/BRM/SMARCA4 Degrader (PROTAC®; IC50 range = 9 - 390 nM in glioma models and cell lines). Comprises a BRG1 inhibitor joined by a linker to a cereblon (CRBN) E3 ligase ligand. Completely degrades BRG1 after 48 and 72 hours in HSJD-DIPG007 and SU-DIPGXIIIP cells and reduces viability of a variety of cancer cell lines including H3K27M glioma, prostate, thyroid and salivary gland.

Degraders (e.g. PROTACs) are bifunctional small molecules, that harness the Ubiquitin Proteasome System (UPS) to selectively degrade target proteins within cells. They consist of three covalently linked components: an E3 ubiquitin ligase ligand, a linker and a ligand for the target protein of interest. Authored in-house, this poster outlines the generation of a toolbox of building blocks for the development of Degraders. The characteristics and selection of each of these components are discussed. Presented at EFMC 2018, Ljubljana, Slovenia
