Biological Activity for TL 13-112

TL 13-112 is a selective anaplastic lymphoma kinase (ALK) Degrader (PROTAC®) (DC50 values are 10 and 40 nM in H3122 and Karpas 299 cells, respectively). Comprises the cereblon E3 ligase ligand Pomalidomide (Cat. No. 6302) conjugated to an ALK inhibitor. Inhibits proliferation of ALK-positive cancer cell lines. Maximum degradation exhibited at 16 h.

Negative control TL 13-110 (Cat. No. 6746), and ALK antibody validated for Simple Western™ (automated Western) instruments and Western Blot also available: Catalog # AF4210.

Degraders (e.g. PROTACs) are bifunctional small molecules, that harness the Ubiquitin Proteasome System (UPS) to selectively degrade target proteins within cells. They consist of three covalently linked components: an E3 ubiquitin ligase ligand, a linker and a ligand for the target protein of interest. Authored in-house, this poster outlines the generation of a toolbox of building blocks for the development of Degraders. The characteristics and selection of each of these components are discussed.
