Neuropeptide FF/AF (NPFF) receptors are a group of Gi/Go-protein-coupled receptors that are divided into two subtypes; NPFF1 and NPFF2. NPFF receptors act through inhibition of adenylyl cyclase and activation of phospholipase C and are coupled to voltage-gated N-type calcium channels. NPFF receptors have been implicated in hormonal modulation, regulation of food intake, thermoregulation and nociception through modulation of the opioid system.

NPFF receptors are distributed across the central nervous system, but each subtype has specific localizations. NPFF1 are found in their highest concentrations in the septum, nucleus of the tractus solitarius and hypothalamus, suggesting their participation in neuroendocrine functions. NPFF2 are found in their highest concentrations in the diencephalon and superficial layers of the spinal cord, suggesting their participation in the modulation of pain. The human genes encoding NPFF1 and NPFF2 receptors are located on chromosomes 4q21 and 10q21-22 respectively.
