Biological Activity for UH15-38

UH15-38 is a potent and selective receptor interacting protein kinase 3 (RIPK3) inhibitor (IC50 = 20 nM). UH15-38 potently and selectively blocks influenza A-induced necroptosis in alveolar epithelial cells in vivo (IC50 = 39.5 nM), and prevents damage from lung inflammation without compromising antiviral adaptive immune responses or impeding viral clearance. Blocks TNFα-induced necroptosis in primary murine embryonic fibroblasts (IC50 = 98 nM) and mouse cell lines in vitro.

Cell Viability Published vs Bio-Techne batch comparison for UH15-38 View Larger

Published vs Bio-Techne batch comparison for UH15-38. Inhibition of necroptosis by UH15-38 in human FADD-deficient Jurkat cells. Cells were treated with 10 ng/ml of human TNF alpha for 24 hr in the presence of the dose range of the inhibitors. Cell viability was determined using CellTiter-Glo assay (Promega). IC50 for the published molecule was 238.2 nM and 160.2 nM for the Bio-Techne batch. Data kindly provided by A. Degterev’s laboratory, University of Tufts.
