ER resident protein, STIM1 and PM resident Orai1 channel were expressed with YFP and CFP fluorescent tags respectively in HEK293 cells and imaged using TIRF microscopy. Basal STIM1 and Orai1 (left) co-clusters to form punctae at PM-ER junctions upon ER store depletion (right) by BHQ (30 µM), leading to store-operated Ca2+ entry through Orai1.
