生物活性 for INF 4E

INF 4E is a NLRP3 inflammasome and caspase-1 inhibitor (Ki = 9.6 μM for caspase-1). Inhibits Nigericin (Cat. No. 4312) or ATP (Cat.No. 3245) induced pyroptosis of PMA-differentiated and LPS-primed THP-1 cells and activates the Keap1-Nrf2 pathway.

We used this product to study whether bacterial component can induce NLRP3-mediated IL-1beta production. To confirm whether this inhibitor works well, we pretreated THP-1 cells with the inhibitor for 1 h, followed by treatment with LPS+ATP for 2 h. The production of IL-1b in culture supernatant was measured by IL-1b ELISA kit. We showed that this inhibitor can significantly inhibit LPS/ATP-induced IL-1b production.
