生物活性 for Cytarabine

Cytarabine is a nucleoside analog of deoxycytidine; inhibits DNA replication by incorporating into DNA (IC50 = 0.04 μM in L1210 and CEM cell lines). Displays no inhibitory effects on RNA synthesis. Causes S phase cell cycle arrest in ML-1 cell lines; cytotoxic in L5817Y leukemia cells. Antineoplastic and antileukemic agent.

Cytarabine, also known as AraC, is an anti-mitotic drug. I have used it to inhibit neurogenesis. To deliver this drug into the brain, I implanted a chronic osmotic minipump with cannula located in the lateral ventricle of mice. The drug was diluted in NaCl 0.9% at 2% concentration (m/v) and delivered for 14 days. After treatment, a proliferative zone of the brain was immunolabeled for the doublecortin marker, which stains new neurons. The image shows that the mouse injected with Cytarabine (RIGHT) displayed no signal of new cells, suggesting that neurogenesis was blocked.
