Biological Activity for Flutax 2

Key information: Flutax 2 is a green-fluorescent taxol derivative; for microtubules labeling in live cells.

Used for: imaging microtubules in live cells, isolated cytoskeletons and microtubule suspensions.

Application: fluorescence microscopy.

Properties and Photophysical Data: The fluorescent label on Flutax 2 is attached by derivatization of the 7-β-hydroxy group of native paclitaxel which permits selective binding of the probe to microtubules. Flutax 2 binds microtubules with high affinity (Ka ~ 107M-1). Flutax 2 binds to α, β tubulin dimers and stabilizes them. Flutax 2 is soluble and pH insensitive and is more photostable than Flutax 1 (Cat. No. 2226). Excitation and emission maxima (λ) are 496 nm and 526 nm, respectively.

Flutax 2 labeled HeLa cells. Flutax 2 was diluted in HBSS and applied to live HeLa cells at a concentration of 2 μM. HeLa cells were incubated for 1 hour at 37°C. After incubation was complete, Flutax 2 was washed off, and fresh HBSS was applied to the cells. Cells were not fixed because Flutax 2 staining is not retained at all after fixation (photobleach). Like Flutax 1, Flutax 2 staining in live cells diminishes very rapidly when exposed to light, so the cells should be exposed to light as little as possible to retain good signal.
