Biological Activity for 5-Ph-IAA

5-Ph-IAA is a selective and potent "Bump & Hole" TAG Degrader (DC50,6h = 17.0 nM, T1/2 = 62.3 min), for use within an auxin-inducible degron (AID2) system. 5-Ph-IAA is an analog (the “Bump”) of natural auxin that induces rapid degradation of a target protein tagged with mAID F-box protein (the "Hole" or mini AID), and recruits a mutant E3 ligase complex, OsTIR1(F74G). The induced degradation is reversible 3 h after washout with fresh medium. 5-Ph-IAA shows rapid depletion of mAID-tagged nuclear and cytoplasmic proteins in proliferating and non-proliferating mammalian cells with no detectable basal degradation, lower cytotoxicity level, increased potency and faster degradation of the reporter in comparison with the conventional AID system. The AID2 system also works in budding yeast and fission yeast, and it can control protein expression in multiple mouse organs in vivo.

5-Ph-IAA-AM (Cat. No. 7893), an analog of 5-Ph-IAA, is also available.

Plasmid vectors for the AAV transfection and CRISPR-mediated knock-in of OsTIR1F74G and mAID tag are available from Addgene.

Targeted Protein Degradation Poster

Targeted Protein Degradation Poster

Degraders (e.g. PROTACs) are bifunctional small molecules, that harness the Ubiquitin Proteasome System (UPS) to selectively degrade target proteins within cells. They consist of three covalently linked components: an E3 ubiquitin ligase ligand, a linker and a ligand for the target protein of interest. Authored in-house, this poster outlines the generation of a toolbox of building blocks for the development of Degraders. The characteristics and selection of each of these components are discussed. Presented at EFMC 2018, Ljubljana, Slovenia

Validating Targets for TPD Using dTAG Poster

Validating Targets for TPD Using dTAG Poster

The dTAG platform offers a generalizable strategy to degrade, in principle, any intracellular protein of interest (POI) and is a useful strategy for exploration and validation of targets. This poster presents a workflow solution for target validation, from custom TAG knock-in cell-lines for your POI, different dTAG Degraders to knockdown you POI, to automated assays for protein degradation using Simple WesternTM instruments. Data are also presented on the use of an antibody that recognizes the TAG domain (FKBP12F36V) for detection of degradation. Presented at TPD Europe, March 2022, London, UK.
